01 · Artiva ends AlloNK: clinical risk becomes a corporate reset · $ARTV

2026-10-27

Artiva is ending AlloNK development, including its planned Phase 3, after reassessing benefit, response durability and safety. One fatal event was considered likely related to fludarabine neurotoxicity; the second was judged unrelated to treatment components, with its cause undetermined. The planned workforce reduction is approximately 94%. Cash, equivalents and investments were about $319.1 million at September 30, a preliminary, unaudited figure.

The shareholder question changes: close-out costs, liabilities and the destination of remaining resources now matter alongside clinical results. Reported cash is not a distributable amount. Strategic alternatives include corporate transactions, asset sales and potentially liquidation, with no guaranteed outcome.

What comes next: orderly trial close-out and strategic review; workforce reductions expected by the first quarter of 2027. Key risk: total costs cannot yet be reasonably estimated and strategic outcomes remain uncertain.

Release / document 1 · Release / document 2 · Hub $ARTV

02 · BEAM-301: two patients stop starch supplementation, with safety in focus · $BEAM

2026-10-27

By August 17, six patients had received BEAM-301 in the open-label Phase 1/2 study for R83C-associated GSDIa. In the 40 mg cohort, two of three reached 15 hours of controlled fasting and stopped starch supplementation. Four of the six patients overall experienced serious adverse events considered treatment-related, including three Grade 3 infusion reactions. Premedication and infusion rates have been modified.

On October 27, Beam presents at ESGCT findings previewed on October 13. Fasting control is a tangible outcome in a disease requiring intensive dietary management. The very small sample makes each patient’s course consequential and requires efficacy and safety to be assessed together.

What comes next: enrollment in the 60 mg cohort and planned FDA discussions. Key risk: durability, reproducibility and tolerability in a broader population.

03 · Merck: sac-TMT extends survival, with a separate Keytruda advance · $MRK

2026-10-26

TroFuse-005, a Phase 3 study in 776 patients with endometrial cancer previously treated with platinum and anti-PD-(L)1 therapy, showed median overall survival of 19.0 versus 12.1 months with chemotherapy: HR 0.68. PFS was 7.2 versus 3.9 months, HR 0.59. Grade 3 or higher treatment-related events occurred in 54.4% versus 41.4%. The BLA is under Priority Review, with an FDA target date of June 15, 2027.

In the separate first-line dMMR KEYNOTE-C93 study, Keytruda monotherapy produced PFS of 38.2 versus 8.4 months; an overall-survival advantage was not statistically established. The trials address different populations and treatment settings.

The useful read-through is a broader range of options along the treatment pathway. For sac-TMT, a survival benefit carries more weight than response alone, but must be weighed against toxicity and the pending regulatory review. The full presentation adds detail to previously announced positive results.

What comes next: BLA review and a fuller assessment of benefit and risk.

04 · Pfizer: an earlier TALZENNA approval and a Phase 3 win for ELREXFIO · $PFE

2026-10-27

On October 26, the FDA approved talazoparib with enzalutamide for metastatic prostate cancer with HRR mutations that is naïve or sensitive to androgen-pathway modulation. This is an actual indication expansion for TALZENNA plus XTANDI.

On October 27, MagnetisMM-32 met its primary PFS endpoint for ELREXFIO in relapsed/refractory myeloma after at least one prior line including anti-CD38 therapy and lenalidomide. Estimated 18-month PFS was 68%; median PFS was not reached and OS remains immature. Grade 3 or higher infections occurred in 27.5% versus 18.1% with comparator regimens.

For TALZENNA, attention turns to adoption in the newly authorized population. For ELREXFIO, Phase 3 strengthens the evidence package, while potential earlier-line use depends on subsequent regulatory steps. An approval and positive trial results remain different milestones.

What comes next: full MagnetisMM-32 details, OS maturation and regulatory initiatives. Key risk: infection management and positioning in an increasingly competitive myeloma market.
